Film

Madhoo started her career with the Tamil drama movie Azhagan as Swapna in 1991. Later she got featured in the Hindi action-romance movie Phool Aur Kaante as Pooja Salgaonkar. The same year, she appeared in the Malayalam comedy movie Ottayal Pattalam as Gopika Varma/ Indu.

In 1995, she has done several movies including Diya Aur Toofan as Aasha, HathKadi as Rani, Jallaad as Gayetri, Ravan Raaj as Geeta Suzie face, and Hum Hai Bemisaal as Marya. She acted the role of Archana Patkar in the 2002 Hindi film Mulaqaat. She played the role of Shakuntala Devi in the Kannada action-drama movie Seetharama Kaylana in 2019.

In 2021, she appeared in the Hindi-Tamil biographical drama film Thalaivi as V.N. Janaki Ramachandran. She did a Telugu movie Shaakuntalam as Menaka in 2022.

Madhoo debuted with the Tamil television series Kaveri as female protagonist in 2001. She got featured in the Hindi series Devi as Goddess Durga on Sony TV in 2001. She appeared in the prominent role of Soundaravalli, a character from the Devadasi community, in the Tamil serial Soundaravalli in 2012.

In 2017, she acted in the Hindi historical-fiction serial Aarambh: Kahaani Devsena Ki as Maharani Sambhavija.
