The legendary Yash Chopra noted Poonam’s talent and cast him in the film Trishul in 1978, after winning the titles of Miss Femina India and Miss Young India in 1977 and, 1978 respectively. She debuted as an actress in 1978 at the age of 16 and gained popularity and focus in the film industry after the achievement of her second film Noorie in 1979. She has appeared in several vernacular films, including Nyay Danda in Bengali, Yuddha Kaanda in Kannada, and Ishtam in Telugu.
In her cinematographic career, she has participated in many television series, shows, and plays, including The Perfect Husband and The Perfect Wife. She has worked in several successful films like Kaala Patthar, Red Rose, Biwi O Biwi, Yeh Toh Kamal Hogaya, Teri Meharbaniya, Karma, and many others. Dhillon was a contestant on Bigg Boss season 3 that broadcast on Colors TV channel in 2009. She ended up second in the spectacle. Her grand return to Indian television in a lead role was with the soap opera Ekk Nayi Pehchan on Sony Entertainment television in 2013.
